Is the motor supposed to be in neutral and runing to check transmission

Hello, 

The engine should be running, and it should be at normal operating temperature. Keep the vehicle in park rather than in neutral. Wipe the dipstick, then reinsert for a moment to get a true reading on the dipstick. You can also add transmission fluid if needed through the dipstick tube. Good luck!
